AP06 Baumeister/Sakata: The 26 S Proteasome: dynamics of the regulatory particle
The 26S proteasome is responsible for the regulated degradation of intracellular proteins. The 2.5MDa complex built of 34 canonical subunits comprises two subcomplexes: The core particle (CP) which executes degradation and one or two regulatory particles (RPs) which prepare substrates for degradation. Recently, we determined the structure of the holocomplex to near-atomic resolution. Using in vitro and in situ approaches developed during the current funding period, we aim for a comprehensive understanding of the conformational changes the regulatory particles undergo during the proteasome's functional cycle.
